Sinopsis:
Charles Schine (Clive Owen), que trabaja como ejecutivo en una agencia de publicidad, es una de tantas personas que acuden cada mañana a su trabajo en Chicago en el tren de las 8:43. Pero un día pierde ese tren y conoce por casualidad a Lucinda Harris (Jennifer Aniston)... A partir de ese momento, su vida dará un giro de 180 grados. Lucinda es encantadora, guapa y atractiva. Aunque los dos están casados y tienen hijos, sienten una atracción irresistible. Las citas para quedar a comer se van convirtiendo en copas después del trabajo y, finalmente, Charles y Lucinda deciden consumar su relación en la habitación de un hotel. Lo que parece una aventura absolutamente perfecta se ve truncada cuando La Roche (Vincent Cassel), un despiadado desconocido, irrumpe en su habitación a punta de pistola. Su relación extramatrimonial se convierte en una pesadilla mucho más peligrosa y violenta de lo que jamás podrían haber imaginado.
